TVCB01 Methods in structural biology, 6 ECTS-points
/Strukturbiologiska metoder/

Advancement level:
C

Aim:
The goal of the course is to give basic knowledge on the use of different methods to study the structure of biological molecules, cells and tissue.

Prerequisites:
Biochemistry (TFKE 25) and Cellbiology (TFBI 10).

Course organization:
Lectures, demonstrations and laboratory work.

Course content:
Preparation of tissue for microscopy: fixation, embedding, cutting, mounting of sections and staining. Construction of the electronmicroscope: different types of electronmicroscopes. Studying of biological tissue in the electronmicroscope. Freeze etching and freeze fracturing. Examination of biomolecules: NMR, x-ray crystallography and circular dicroism. Biological markers: Immunocytochemical methods. How to synthesize monoclonal and polyclonal antibodies. Autoradiography for light microscopy and electron microscopy. Quantitative analysis: Cytometry, including flow cytometry.
