Aim:
To provide the student with practical ability and theoretical knowledge about cultivation of microorganisms and animal cells. The course is closely connected to previous courses in cell biology and molecular biology in the program and intend to bridge the gap between biology and engineering. The course prepares the student for academic research, industrial research and development and production.

Prerequisites:
TRTE01 Large-scale biological processes or equivalent.

Course content:
Microbial biotechnology, scope and techniques. Microorganisms as living factories for macromolecules. Production of proteins in bacteria. Production of proteins in yeast. Production of synthetic vaccines, insecticides, enzymes and polysaccharides. Microorganisms in plant biotechnology. Biomass to fuel, ethanol. Metabolites from microbes; amino acids and antibiotics. Environmental applications. Using mammalian cell cultures for antibodies, biopharmaceuticals and gene therapy. Cultivation technology in practice and theory. To setup a lab fermentation. Strain preservation and inoculum preparation. Measurement methods. Instrumentation and control. Sterilization. Media preparation for animal cell cultures. Scaling up of mammalian cell cultures. Cell line preservation. Analysis methods. Lab exercises. Project work: 3-4 weeks project with 2 students. Essential features of bioprocesses are covered. Examples: fed-batch fermenation with recombinant strain. Studies of medium effects on metabolism and physiology. Cultivation of CHO cells. Affinity chromatography.
