Aim: The course should provide theoretical and practical knowledge in molecular genetic technologies and their use in the biomedical field. The course should also illuminate the use of databases covering genetic information.Prerequisites: Biochemistry (TFKE 25) and Cell Biology (TFBI 10).Course organization: The course consists of lectures, seminars and laboratory work.Course content: DNA: Control of gene expression in eukaryotic cells. DNA damage and repair. Medical genetics and the HUGO project. Genetic variation, polymorphism and mutation. Cytogenetics. Basic gene technologies: Restriction enzymes and RFLP. Separation of nucleic acids. Immobilization on membranes (Northern and Southern blotting). Labeling and hybridization. Nucleotide sequencing. Isolation of genes (cloning) and use of vectors. Site-directed mutagenesis. PCR. Use of computers in molecular genetic work (sequence databases). Applications of gene technologies in biomedicine: Clinical diagnostics, linkage analysis, prenatal diagnostics and genetic counseling. Applications in forensics.
